3rd Gen-GP50 (better then Kato's) Yeah, that was a great engine 20 or so years ago. I had one but sold it when I decided to model the transition era. However, Kato or someone else sure could bring out a new one out with the latest frame technology.
C415, SD70ACe, F9, SW1500/1000, GP15-1, a D&RGW L-132 2-8-8-2......... I tossed that last one in to see if you're paying attention!
The only locomotive that I would probably drop everything and order right now is the GP50. I can stand on my existing roster and have a pretty convincing representation of the prototype I'm trying to model, but the GP50 is definitely a hole I would like to fill and preferrably with a new release. Others might include a refreshed C30-7, the Dash 8-40C, chop-nose GP7's, and CF7's. Could also use another run of BNSF Dash 9's and an SP AC4400.
Hmmm... hard question. Here's my list. I know some are coming soon, but at the moment, I still consider "soon" to be missing. F45, FP45, SD70ACe, ES44AC, SDL39 (gotta throw that out there because it's a favorite), SW1500, SW1, GP50, GP15 (all versions), CF7
Unbelievably that list includes the S-4 , AS 616 , and a valve gear equipped 0-6-0 . Then there is the H-20-44 SW-1 GE 44T Heisler and about a gazillion more .
Good grief, too many to mention. How about some center cabs, any size will do. Electrics of any kind, we have almost none. Heislers, Climaxes, and three-truck Shays, oh my! Camelbacks, anyone? Not to mention better versions of many steamers still only available as old tooling from Bachmann et al.
I'll add to my list: a UP FEF-3 4-8-4 Northern type, as well as a heavy Mountain. As for engines that need re-run: SD45, C30-7/U30C from Kato I'll voice my desire for SP AC4400's as well. No self-respecting Colorado mountain coal hauler can be without Espee ACeez!

I will second David Smith's call for more variety of good steam. The Shay that Atlas put out is great. So is Kato's Mikado. I would love to see some Heislers and Climaxes and even a couple of Willamettes, or at least detailing packages so that I can kitbash a Shay into a Willamette since they are nearly the same thing. Some of these are probably available in brass, but staying married is more important to me than collecting brass locomotives. I would love to see more RTR stuff in Nn3 that isn't $500+. Apart from that, I would love to see some more speeders and M-O-W equipment in N scale.
In terms of popularity Alco S2 Diesel - by a long shot over any other diesel 4-6-0 Steamer - also by a long shot over any steamer In terms of convenience for my tastes Alco RS27 Baldwin Centercab GE U25C or U28C EMD SDP40 or SDP45 or NW5
GE U-25C, U-28C, U33C and the U36C, 2-8-8-4 Yellowstone, AS616, U28B, Alco S-2, S-3 and S-4, SDL39, more variations of the SD40-2. Need these to fill out my NP and MILW rosters.
The last 3 choices you made show you're a GN modeler! I would LOVE to have SDP45's! The NW5 is so rare and funky, it's cool.
Since I model modern CSX, I could realy use a few C40-8, CW40-8, AC60CW, SD70ACe, ES44DC, MP15AC and T, GP15, later phase GP38-2 with nose headlight, a correct DCC GP60, and a few FEC SD70M-2's. As for a steamer to run my excursion train, I would also not mind getting a good running plastic C&O Alleghenies, just the thing to make give a UP Big Boy an inferioty complex.:teeth:
